PEG-12 DIMETHICONEDermally administered PEG-12 dimethicone (100%; 0, 50, 100 or 200mg/kg) was not embryotoxic or teratogenic toNew Zealand White rabbitstreatedon days 6 through 18 of gestation.30No adverse effects were observed in mean maternal body weights, feed consumption,or liver weights of the treated rabbits. No differences were observed in the number of implantation sites, number of live fetuses per litter, mean litter size, fetal body weightsor crown-rump length between the control and treated groups. The incidence of resorption among the total fetal population ofrabbits treated with test material was similar to that of the controlgroup. No single alteration was observed inanyofthe treated litters which wasdifferent from the controlgroup. No treatment related signs of toxicity or behavioral changes were observed in any of the pregnant rabbits. Three rabbits in the 50-mg/kg/d group, 1 in the 100-mg/kg/dgroup, and 3 in the 200-mg/kg/dgroup died during the study. Some pregnancies were terminated because of Pasteurella multocidainfection, a known abortifacient in rabbits.Waterserved as the control. One pup in the treatment group displayed clubbing of the extremities, partial acranius, and an umbilical hernia. The 24-hsurvival of fetuses, abnormalities in the fetuses, and number of abnormal fetuses were similar across bothcontrol groups(administered the vehicle or nothing). No dermal effects were reported for the dams. Subsequent testing of the test substance demonstrated that it was neither embryotoxic nor teratogenic when dermally administered to New Zealand whiterabbits at dosesof 50, 100, or 200 mg/kg (data not provided).
